Set thermostat at 280F. Bring mixture to a boil and boil for 5 minutes, stirring constantly. Turn off heat. Add marshmallows, peanut butter, and vanilla. Stir until marshmallows and peanut butter are melted and blended. Pour into a buttered 8-inch square pan.
